The invention relates to a polypeptide sequence specifically bonded with poultry bursal virus VP2 protein, and an application of the polypeptide sequence, and belongs to the field of molecular biology. Through computer-aided design, based on the crystal structure of the poultry bursal virus VP2 protein, through a molecular docking virtual screening technique, polypeptide ligands which are optimalin target protein binding mode and affinity are searched in a virtual polypeptide database, and the polypeptide sequence is SERWDN namely IBDVL9-15. Expressed and purified IBDV VP2 protein is subjected to ELISA and localized surface plasmon resonance experiment, the result indicates that IBDVL9-15 polypeptide can have favorable binding capacity with IBDV VP2 protein, and the designed polypeptide can be used for performing fast detection as well as separation and purification on IBDV VP2 antigens.
