[0018] 1) Cultivation of test nematodes and preparation of nematode suspension

[0019] The pine wood nematodes tested were provided by Jiangsu Key Laboratory of Pest Invasion Prevention and Control, Nanjing Forestry University, and were isolated from black pine trees in Lianyungang, Jiangsu Province. The pine wood nematode was cultured with Botrytis cinerea (Botrytis cinerca). The nematodes were inoculated on a PDA plate covered with Botrytis cinerea, and placed in an incubator at 25°C for cultivation until the hyphae of Botrytis cinerea disappeared and the nematodes covered the plate. The nematodes were separated and collected by the Baermann funnel method, and then centrifuged, sterilized and concentrated to prepare a nematode suspension of about 20 nematodes / μL.

Abstract

The invention discloses pine wood nematode-killing Bacillus cereus, which is classified and named as Bacillus cereus NJSZ-13, and preserved in China type culture collection center, the preservation number is CCTCCM2016660, the preservation date is November 20, 2016, and the preservation address is Wuhan University in Wuhan in China. The Bacillus cereus NJSZ-13 isolated from bodies of pine trees has better competitive ability and colonization ability, and sterile fermentation broth and bacteria aqueous suspension of the Bacillus cereus NJSZ-13 both have strong nematicidal activity on pine wood nematodes. By experiments, 36h after the treatment, the killing rate of the sterile fermentation broth on the pine wood nematodes can reach 100%, bodies of the pine wood nematodes can be digested, 48h after the treatment, the killing rate of the bacteria aqueous suspension on the pine wood nematodes can reach 86.5%, and the pine wood nematode-killing Bacillus cereus has good practicability.
