preparation example Construction

[0023] The invention provides a method for preparing a biochip based on a three-dimensional structure nanoarray, the method comprising the following steps:

[0024] (1) Treat the gold film with a silane solution containing mercapto;

[0025] (2) Treating the gold film treated in step (1) with an acid solution;

[0026] (3) Pasting the anodized aluminum film with through holes on the gold film treated in step (2);

[0027] (4) Use the sol-gel method to impregnate the gold film obtained in step (3) in silicon tetrachloride (SiCl 4 ), n-hexane, a mixture of n-hexane and methanol, ethanol and water (this process can be recycled several times);

[0028] (5) Treating the gold substrate obtained in step (4) with an acid solution to remove the aluminum oxide film.

[0029] According to the present invention, in step (1), the gold film is a glass substrate material with a gold layer on its surface. In the present invention, a layer of gold with a thickness of 35-50 nm, preferably 40-4

Embodiment 1

[0041] This example is used to illustrate the preparation of a biochip with a three-dimensional silica nanotube array structure using the method of the present invention.

[0042] (1) Preparation of gold film: place a K9 glass sheet in H with a volume ratio of 7:3 2 SO 4 (98% by weight) and H 2 o 2 (30% by weight) mixed solution, washed for 1 hour, and then rinsed with deionized water. The cleaned K9 glass sheet is vapor-deposited with a layer of 40nm thick gold by magnetron sputtering, which is the gold substrate used in this embodiment;

[0043](2) Treatment of gold substrate with mercaptosilane solution: Prepare 20 mmol of (3-mercaptopropyl)trimethoxysilane solution with methanol. Immerse the gold substrate obtained through step (1) in the above solution for 3 hours.

[0044] (3) Acid solution treatment of the gold substrate: use 0.1 mol hydrochloric acid solution to treat the gold substrate after step (2), and the treatment time is 10 hours at room temperature;

Abstract

The invention discloses a preparation method and applications of a three-dimensional structure-based nanoarray biochip. The method comprises the following steps: (1) treating a gold film through silane solution containing sulfhydryl group; (2) treating the gold film, treated by the step (1), through acid solution; (3) attaching an anodized aluminum thin film of a through hole onto the gold film treated in the step (2); (4)dipping the gold film, obtained in the step (3), into SiCl4, n-hexane, hexane-methanol mixed solution, ethanol and water sequentially (circularly carrying out the processes); (5) treating a gold substrate, obtained in the step (4), through acid solution. According to the method, a nanotube (column) array with the three-dimensional structure is easily prepared on the substrate, the size of the tube (column) is controllable, the three-dimensional structure-based nanoarray biochip can be used as a biochip, and the high-sensitivity and label-free biodetection can be carried out.
