[0020] Thirty ICR adult male mice (animal production license: SCXK (Su) 2017-0007), weighing 25-29 g, were purchased from the Comparative Medicine Center of Yangzhou University. They were adaptively raised in the experimental animal room of the Department of Integrated Traditional Chinese and Western Medicine of Yangzhou University for one week. The ambient temperature of the animal room was 20-26°C and the humidity was 40-60%.

[0021] After the adaptive feeding, the baseline body weight measurement, open field test, sucrose water preference test, tail suspension test, forced swimming test and feces traits were completed.

Abstract

The invention relates to the technical field of depression research, in particular to a modeling method for mice with diseases of depression and liver-stagnation and spleen-deficiency syndrome. Healthy mice are continuously stimulated for 28 days, each type of stimulation is averagely used for 4 times, and the stimulation types are as follows: water forbidding and food interruption stimulation for24 hours, tail clamping stimulation for 1 min, ice water swimming stimulation at 10 DEG C for 1 min, horizontal shaking continuous stimulation at the frequency of 1 time/s for 5 min, lamplight irradiating stimulation for 24 h, saturated water wet padding stimulation for 24 h or continuous stimulation by inclining a mouse cage by 45 degrees for 24 h; from the fifteenth day to the 28th day of continuous stimulation, 0.5 mL of rheum officinale powder aqueous suspension with the concentration of 15% (wt) is further perfused into stomachs of the mice every day. The obtained model mice have the symptomatic expression of depression and liver-stagnation and spleen-deficiency syndrome, and can be used for further research on pathogenesis and treatment of depression and liver-stagnation and spleen-deficiency syndrome.
