[0028] The test materials were two-year-old cuttings of two North American holly varieties Ilex verticillata 'Oosterwijk' (Oosterwijk) and Ilex verticillata 'Apollo' (Apollo) with the same growth. In late March, all the seedlings were transplanted into plastic pots of the same specification with a diameter of 16 cm and a depth of 15 cm, and one plant was planted in each pot. Each pot contains 1.7kg of cultivation soil, which is made by mixing garden soil and nutrient matrix at a ratio of 3:1. After transplanting, the seedlings are slowly seeded in the greenhouse for 2 months, and 80%-85% of the normal soil field water holding capacity is maintained during the growth process for daily watering without fertilization.

Abstract

The invention provides a research method for increasing the oxidation resistance of Ilex verticillata. The method includes: selecting the biennial cuttage seedlings of two Ilex verticillata varieties,and performing acclimation in a greenhouse for 2 months; selecting 18 cuttage seedlings of each of the two varieties, separating each variety into two groups, spraying melatonin to one group, and spraying equal-amount clean water to the other group; performing identical drought stress treatment at the same time; collecting fresh leaves immediately after the treatment, on the seventh day after thetreatment and on the thirtieth day after the treatment, quickly freezing the collected leaves in liquid nitrogen, and preserving the frozen leaves in refrigerator of ultralow temperature of minus 80DEG C; measuring superoxide dismutase and peroxidase activity and malondialdehyde content. The research method has the advantages that by applying external melatonin to perform treatment, the influence of the melatonin on the oxidation resistance of the Ilex verticillata cuttage seedlings under drought stress is comparatively analyzed; the research results can lay a foundation for the research ofthe Ilex verticillata physiological mechanism responding to the drought stress and measures for increasing the drought resistance of the Ilex verticillata and the popularization of the Ilex verticillata in drought regions.
