Embodiment 1

[0044] A preparation method of squid ink preparation with breast cancer chemotherapy synergistic activity, comprising the steps of:

[0045] (1) Take the frozen squid ink sac and thaw it at 4°C, take out the ink, add 1 times the volume of ice-cold phosphate buffer saline PBS, the pH of PBS is 7.2, and the concentration is 0.01M. 5s, 10s interval, 20 times;

[0046] (2) The ultrasonically treated suspension was stirred and soaked at 4°C for 48 hours, then centrifuged at 4°C for 60 minutes, and the speed of the centrifuge was 8000rpm;

[0047] (3) Remove the precipitate from the centrifuged liquid, add papain to the supernatant to a concentration of 0.15%, and incubate in a water bath at 55°C for 60 minutes;

[0048] (4) Heating in a water bath at 100°C for 20 minutes, after cooling, centrifuge at 4°C for 60 minutes, and the centrifuge speed is 8000rpm;

[0049] (5) Remove the precipitate from the above centrifuge, add 1 / 4 volume of chloroform and n-butanol mixture to the super

Abstract

The invention relates to a preparation method and application of a sepia preparation with breast cancer chemotherapy enhancing activity and belongs to the technical field of biological engineering; the preparation method includes the steps of adding PBS into sepia, grinding well, ultrasonically treating, centrifuging to remove precipitate, adding papain into supernate, incubating by water bath, cooling, centrifuging to remove precipitate, adding chloroform and n-butanol mixed liquid into the supernate, mixing well and then centrifuging, taking yellow supernate, repeating mixing and centrifuging 2 to 3 times, combining yellow supernates, concentrating with a rotary evaporator, adding absolute ethyl alcohol into concentrate, standing and then centrifuging, collecting precipitate, spray-drying and then crushing into fine powder, and filling hard capsules; the sepia preparation of the invention can inhibit breast cancer cell activity and metastasis, can also synergically enhance cisplatin by inhibiting cisplatin-induced cell autophagy to promote withering of triple negative breast cancer cells, thereby enabling killing action, and can be used in auxiliary therapy for tumor chemotherapy.
