AI Technical Summary

Benefits of technology

This patented technology allows creatures called Achromobacters (a type of gram negative bacillus) that have been found naturally on rocks or plants like chambeads to break down hardy substances such as amber nitrogen fertils. These microorganisms use hydrolysis energy derived from water molecules during their growth process to dissolve these compounds into liquids instead of burning them away at once. They also reduce harmful chemicals released over longer periods compared to traditional methods used alone. Overall, this innovation provides technical benefits including efficient decomposition of challenged organoid materials without causing environmental concerns.

Problems solved by technology

The problem addressed in this patents relates to finding ways for effectively removing lignocellulosic material from water without causing environmental concerns or requiring expensive equipment like traditional processes involving acidifying agents.

[0022] Achromobacter of the present invention ( Achromobacter sp. ) The separation and domestication process of CCZU-WJ6 is as follows:

[0023] 1) Sample, the sample is rotten wood in nature;

[0024] 2) Medium

[0025] Primary screening medium: alkaline lignin medium. Alkaline lignin medium (g / L): Alkaline lignin 2, ammonium sulfate 1.33, magnesium sulfate 0.5, potassium dihydrogen phosphate 1, disodium hydrogen phosphate 0.2, agar 20, pH7.0.

[0026] Re-screening medium: aniline blue solid medium. Aniline blue solid medium (g / L): peptone 10, beef extract 5, sodium chloride 10, aniline blue 0.08, pH 7.0.

[0027] Enrichment medium: LB medium. Enrichment medium (g / L): peptone 10, yeast extract powder 5, sodium chloride 10, pH 7.0.

[0028] 3) Primary screening, secondary screening and fermentation culture operation

[0029] Primary screening: put 5 g of rotten wood i

Embodiment 2

[0034] Example 2: Preparation of wet cells of Achromobacter sp. CCZU-WJ6

[0035] Prepare the components of the activation medium according to the following concentrations: yeast extract powder 5g / L, peptone 10 g / L, NaCl 10 g / L, pH natural, sterilized at 121°C for 20 minutes, cooled after sterilization, and inserted into the slope Seed (the achromobacter that prepares in embodiment 1 ( Achromobacter sp. ) CCZU-WJ6), cultivated for 1 day at 30°C with a shaker speed of 250r / min, and used it as an activation culture seed solution.

[0036] Prepare the components of the fermentation medium according to the following concentrations: sucrose 20 g / L, peptone 10 g / L, NaCl 10 g / L, pH 7.0, sterilized at 121°C for 20 minutes, cooled after sterilization, according to the volume ratio of inoculum 10% was inoculated with the activated culture seed solution, and cultured for 1 day at a temperature of 30°C and a shaker speed of 180 r / min to obtain a fermentation culture solution

Embodiment 3

[0037] Example 3: Preparation of wet cells of Achromobacter sp. CCZU-WJ6

[0038] Same as Example 2, the difference is that the components of the fermentation medium are prepared according to the following concentrations: yeast extract powder 5 g / L, peptone 10 g / L, NaCl 10 g / L, pH natural.

Abstract

The invention discloses an achromobacter sp. for degrading lignin and application of the achromobacter sp. in lignin-containing wastewater treatment, and relates to the technical field of industrial wastewater treatment. The achromobacter sp. CCZU-WJ6 for degrading lignin is preserved in the China General Microbiological Culture Collection Center (CGMCC), and the preservation number of the achromobacter sp. is CGMCC NO.19017. The strain can be applied to degradation of industrial lignin-containing wastewater, and particularly, when the lignin-containing wastewater is degraded, the applicable degradation culture conditions are as follows: the temperature is 20-40 DEG C, the pH value is 4-9, and the concentration of treated lignin is greatly reduced. Compared with the traditional method, thestrain and the application method of the strain have the advantages of high treatment efficiency, wide application range, simple operation, low equipment requirement and obviously reduced operation cost, and solve the problems of complex procedure and carbon source requirement in fungus treatment of industrial lignin-containing wastewater.
