AI Technical Summary

Benefits of technology

This patented technology allows us to measure how well certain proteins called histones have gone through their chemical changes over time without being affected by any illnesses like depression or neurologism. It makes possible accurate detection even if we don’t know what kind of damage they cause beforehand. Additionally, this technique could help predict whether an individual will develop degenerative brain diseases later on after having them undergo treatment.

Problems solved by technology

Technologies aim towards improving healthcare systems through effective therapy strategies involving administering agents called chaperones to improve symptoms caused by certain conditions including autism spectrum disorders (ASPD) and Parkinson’s syndrome. By measuring changes in levels of specific proteins involved in neuronal cell membranes, research groups were conducted into studying how individuals develop this pathology during their lifetime. Current imaging techniques involve X ray fluorescent analysis, computer tomographic images, ultrasonics, etc.; however, none provides precise quantitative assessments of change over time nor making them accessible without costly equipment. Therefore, there remains room for improvement in providing reliable ways to accurately determine if a cognition issue addressed by existing assays may lead to treatment failure.

[0100]Hereinafter, the present invention is described in more detail based on Examples. Examples described below illustrate examples of typical Examples of the present invention and the scope of the present invention should not be narrowly interpreted by Examples.

(1) Serum Sample

[0101]Serum samples of 20 AD (Alzheimer's disease) samples, 20 MCI (mild cognitive impairment) samples, 20 NDC (subject having a normal cognitive function matched with the AD patients in the age and the sex, non-demented control) samples, and 10 ODD (Other demented disease) samples were used. Furthermore, for the ODD, 10 serum samples of each of Dementia with Lewy Bodies and Frontotemporal dementia were used.

(2) Method

[0102]475 μl of 0.1% trifluoroacetic acid (TFA) was added to 25 μl of each serum, and then boiled at 100° C. for 15 minutes. Thereafter, in order to collect peptides having a molecular weight of 10,000 or less, ultrafiltration was performed using YM-10 of Millipore Corporation.

Abstract

This invention provides a method for detecting cognitive disfunction diseases including mild cognitive impairment and Alzheimer's disease using a protein and a peptide of the protein different in the presence level in subjects having a normal cognitive function and patients suffering from cognitive disfunction diseases and a biomarker for detecting cognitive disfunction diseases including mild cognitive impairment and Alzheimer's disease containing the protein and the peptide. This invention is a biomarker for diagnosing cognitive disfunction diseases containing a prothrombin precursor protein of SEQ ID NO: 1 or a peptide THRB containing the amino acid sequence represented by SEQ ID NO: 2 which is a peptide of the protein, a diagnosis method for cognitive disfunction diseases using the biomarker, an antigen peptide represented by SEQ ID NO: 3 for creating a THRB peptide specific antibody to be used in the diagnosis method, and a cognitive disfunction disease diagnosis kit containing the THRB peptide specific antibody.
