The method comprises the steps of S1, sterilizing seeds; S2, preparing a matrix solution; S3, filling the seeds; and S4, refrigerating the seeds. According to the invention, duringrefrigeration, the cell activity of the seeds is reduced, so that the seeds can be stored for a long time; and matrix layers wrapping the outer walls of the seeds can be solidified into solid, and thematrix layers can play a role in external protection during refrigeration, so that the seeds are prevented from directly making contact with liquid nitrogen to be frostbitten, and the survival rate of the seeds is increased.
