Example Embodiment

[0030] Example

[0031] Preparation method of electrochemical immune sensor, preparation process figure 1 As shown, the specific steps include:

[0032] (1) Activation of screen printing carbon electrode (SPCE): Pipette 100 μL of 0.2 M droplets dropped on the surface of the electrode with a pipette, activate the circulating voltammetry, the scan potential range is -1.5 to 1.5V, scan speed It is 0.1V / s, scanning 10 laps, and the circulation voltammetry is stable to indicate activation, and the deionized water is rinsed, and the room temperature is dry.

[0033] (2) Electrodeposition nano gold particles (AUNPS): A 150 μl of mass fraction is 0.15% by a pipette to activate the surface of the working electrode, performing constant potential deposition, deposition potential to -0.2. V, deposition is 2400s, deionized water is rinsed, and it is dry at room temperature;

[0034] (3) Fixture of protein A: PBS dilution of 10 μl of 20 μg / mL of protein A by a pipette in step (2) depositin

Abstract

The invention discloses an electrochemical sensor as well as a preparation method and application thereof. The electrochemical sensor sequentially comprises an activated screen-printed carbon electrode, gold nanoparticles and protein A from bottom to top, norovirus capsid protein monoclonal antibodies are modified on the protein A, and non-specific sites on the protein A are closed sites. The preparation method comprises the following steps: activating the screen-printed carbon electrode, sequentially depositing the gold nanoparticles, modifying protein A, modifying a norovirus capsid protein monoclonal antibody, and closing a non-specific site to obtain the electrochemical sensor. When the electrochemical sensor disclosed by the invention is used for quantitatively detecting the human norovirus capsid protein, the linear detection range is 1-10ng/mL, and the detection limit is 0.635-1.0 ng/mL. The electrochemical immunosensor is convenient to operate, low in cost, high in sensitivity and suitable for rapidly detecting the norovirus on site.
