Problems solved by technology

[0005] At present, there is no specific drug approved for clinical treatment of ZIKV infection or a vaccine to prevent ZIKV infection. In order to resist the potential risks brought by ZIKV, it is urgent to develop small molecule inhibitors that can effectively inhibit ZIKV infection

[0031] Example 1, 6-Thioinos ine can inhibit the replication of ZIKV

[0032] 1. Preliminary screening of 50μM small molecule compounds that can inhibit ZIKV replication by ELISA

[0033] The enzyme linked immunosorbent assay (ELISA) was used to screen 179 small molecule compounds at a single concentration of 50 μM. The steps are as follows:

[0034] (1) Prepare cells: in 100 μL of DMEM medium containing 10% FBS, 1% PS (PS: penicillin-streptomycin mixed solution, Shanghai Biyuntian Biotechnology Co., Ltd., product number: C0222)), Vero cells were plated with 10 4 Cells / well were seeded in 96-well plates at 37°C, 5% CO 2 overnight in the incubator.

[0035] (2) Virus infection: ZIKV virus was diluted to 100 TCID50 using DMEM medium containing 1% PS to obtain virus dilution. Cells were infected with 50 μL of virus diluent for 1 h, and 3 cell wells were reserved without virus infection and used as blank control.

Abstract

The invention discloses an application of 6-Thioinosine in inhibition of ZIKV (Zikov Institute of Kovary) replication. The invention finds that 6-Thioinosine can inhibit the replication of Zika virus in cells. According to the invention, an enzyme-linked immunosorbent assay is adopted to obtain that the EC50 value of 6-Thioinosine for inhibiting the replication of ZIKV in the cells is 18.82 + / -4.66 [mu] M. According to the invention, the EC50 value of 6-Thioinosine for inhibiting the replication of ZIKV in the cells is 18.82 + / -4.66 [mu] M; the EC50 value of the 6-Thioinosine for inhibiting the replication of ZIKV in cells, which is obtained by adopting an indirect immunofluorescence experimental method, is 16.69 + / -7.89 [mu] M; the cytotoxicity of the 6-Thioinosine is detected by adopting a CCK-8 method, the CC50 value is 112.2 [mu] M, and the SI index (CC50 / EC50) is 5.962. The invention provides a scientific basis for research and development of medicines for effectively inhibiting ZIKV.

Description

technical field [0001] The invention relates to the application of 6-Thioinosine in inhibiting the replication of ZIKV in the field of biomedicine. Background technique [0002] Zika virus (ZIKV) is a mosquito-borne virus of the Flaviviridae family, genus Flavivirus, which was first isolated in 1947 from a sentinel monkey in the Zika forest of Uganda. ZIKV is a single-stranded positive-stranded RNA virus with a genome size of about 10.8 kb, and its genome encodes 3 structural proteins and 7 non-structural proteins. Among them, the C-terminus of the non-structural protein NS5 has RNA-dependent RNA polymerase (RdRp) activity, which is involved in the replication of the viral genome and plays an important role in the life cycle of ZIKV. [0003] ZIKV is mainly transmitted by mosquito vectors, but can also be transmitted vertically through the placenta (Brasil et al., 2016), sexually (Foy et al., 2011) and blood (Bierlaire et al., 2017). The main vectors of mosquito-borne tran...
